| I. Background | |
| cAMP 反應元件結合蛋白 (CREB) 位于細胞核內，是一種轉錄因子，在 Ser133 位點(diǎn)被不同的受體活化蛋白激酶（如蛋白激酶 A (PKA)、鈣調蛋白依賴(lài)性蛋白激酶 (CaMK)、絲裂原活化蛋白激酶 (MAPK) 和其他激酶磷酸化后，與靶基因啟動(dòng)子的 cAMP 反應元件 (CRE)結合。CRE 是 CREB 的主要結合位點(diǎn)，負責激活 CREB。CRE 是許多細胞外和細胞內信號通路的焦點(diǎn)，包括 cAMP、鈣、GPCR（G 蛋白偶聯(lián)受體）和神經(jīng)營(yíng)養因子。cAMP/PKA 信號通路對許多生命過(guò)程和生物體至關(guān)重要。在 cAMP/PKA 信號通路中，CREB 通過(guò) PKA 的磷酸化被激活，并與 CRE 結合，一般基序為 5'-TGACGTCA-3'。由于 CRE 是 cAMP/PKA信號通路的調節劑，因此可以研究各種抑制劑的作用。 | |
| II. Description | |
CRE-Luc HEK293 報告基因細胞是由 CRE 調控并表達 Luc 熒光素酶報告基因的 HEK293 細胞。Forskolin 是天然存在的直接腺苷酸環(huán)化酶（AC）/cAMP/CREB 的 激活劑，原理見(jiàn)下圖所示。 Figure 1：CRE-Luc HEK293 原理示意圖 |
