| I. Background | |
| 補(bǔ)體 C5a 是由補(bǔ)體系統(tǒng)經(jīng) C5 轉(zhuǎn)化酶裂解 C5 產(chǎn)生的強(qiáng)效炎癥介質(zhì),作為核心過(guò)敏毒素 通過(guò)結(jié)合受體 C5aR1/C5aR2 驅(qū)動(dòng)中性粒細(xì)胞趨化、氧化爆發(fā)及炎性因子(IL-6/TNF-α)釋 放,在膿毒癥中誘發(fā)“細(xì)胞因子風(fēng)暴”,在自身免疫病(如類風(fēng)濕關(guān)節(jié)炎)中促進(jìn)組織侵 蝕,在腫瘤微環(huán)境募集髓系抑制細(xì)胞(MDSC)介導(dǎo)免疫逃逸。 C5aR1(CD88)是補(bǔ)體過(guò)敏毒素 C5a 的高親和力 GPCR,主要表達(dá)于髓系細(xì)胞(中性 粒細(xì)胞、單核細(xì)胞、肥大細(xì)胞),通過(guò)激活 Gi/Gq 蛋白-PI3K/MAPK 通路驅(qū)動(dòng)炎癥級(jí)聯(lián)反 應(yīng)。 |
