| I. Background | |
| 生長(cháng)分化因子 15（GDF15），也稱(chēng)為巨噬細胞抑制性細胞因子 1（MIC-1），屬于 TG F-β超家族蛋白。GDF15 是一種內分泌激素，主要參與細胞生長(cháng)、分化和組織修復。GDF1 5 作為一種炎癥標志物，在腫瘤、缺血性疾病、代謝紊亂和神經(jīng)退行性過(guò)程的發(fā)病機制中 發(fā)揮作用。 GFRAL（GDNF-family receptor α-like），也被稱(chēng)為 GRAL，屬于膠質(zhì)細胞源性神經(jīng)營(yíng) 養因子家族α樣受體。GFRAL 是一種僅在后腦中表達的跨膜蛋白，需要輔助蛋白 RET 的幫 助才能進(jìn)行信號傳導。當 GDF15 與 GFRAL 結合時(shí)，RET 將自磷酸化并導致下游信號通路 （如 PI3K-AKT 和 PLC-PKC）的激活，從而調節許多生理過(guò)程，包括影響食物攝入和導致 體重減輕。抑制 GDF15-GFRAL 信號通路則能夠增強免疫系統對實(shí)體瘤的殺傷，并能夠治 療或預防癌癥惡病質(zhì)。 |
